摘要
为研究小麦茎秆伸长过程中的分子调控机制,以郑麦9023为材料,采用Affymetrix小麦基因芯片比较了小麦茎秆拔节前期及后期基部茎节组织的基因表达谱。通过对拔节后期和前期的数据比较,以共同上调表达2倍以上的基因为标准,找出了参与小麦茎秆伸长调控的候选基因172个(172个探针组);利用Affymetrix的NetAffx Analysis Center查询和NCBI的BLASTx分析,其中有29个基因已注释了功能;利用MIPS数据库将注释基因分为6类,包括转录、糖代谢、细胞组分生成、植物光合作用、病程相关蛋白基因及钙离子结合蛋白。
To investigate the molecular regulation mechanisms of wheat stalk elongation,the transcription profile later base stalk elongation and control early base stalk elongation mechanisms of wheat cultivar "Zhen9023" were comparatively studied using the Affymetrix wheat GeneChip.Analysis of the microarray data showed that 172 genes(172 probe sets) were up-regulated exceeding 2 folds.Through the sourcing annotations directly from Affymetrix website in tandem with BLASTx analyzing using the Affymetrix probe set sequences,29 candidate genes were annotated.The functional classification of annotated genes were performed based on literature retrieval of the functional categories described in the MIPS database and the 29 annotated genes were classified into 6 functional categories,including transcription,glycometabolism,biogenesis of cellular components,photosynthesis,pathogenesis-related protein gene,Ca2+-binding protein.
