摘要
采用水蒸气蒸馏法(SD)、索氏提取法(SE)及超临界CO2萃取法(SFE-CO2)提取羌活种子挥发油,利用GC-MS联用技术对所得挥发油进行定性定量分析。同时,采用DPPH法和FRAP法对SFE-CO2法所提挥发油的抗氧化活性进行评价,并与典型的合成抗氧化剂BHT进行比较。结果表明,不同方法所提挥发油在外观、得率、化学成分组成及含量上均存在一定差异,采用SD法、SE法、SFE-CO2法所提挥发油的得率分别为2.3%、12.9%、7.6%,分别鉴定出35、50、55种成分,占各自挥发油总量的比例分别为94.64%、99.93%、99.98%。SFE-CO2法所提挥发油清除DPPH自由基的IC50约为1.9mg/mL;其FRAP值随浓度的增大而增大,线性回归方程为y=0.1958x-0.0128(R2=0.9973)。羌活种子挥发油具有一定的抗氧化活性,且呈现明显的量效关系,但较合成抗氧剂BHT弱。SFE-CO2法所提挥发油能更真实、全面的反映药材中的化学成分。
The essential oil from seeds of Notopterygium incisum Ting ex H. T. Chang was extracted by steam distillation (SD), Soxhlet extraction (SE) and supereritical CO2 extraction (SFE- CO2), and qualitatively and quantitatively analyzed by GC-MS. The antioxidant activities of the oil extracted by SFE-CO2 were assessed by 2, 2'-diphenyl-l-pierylhydrazyl radical scavenging method (DPPH) and ferric reducing/antioxidant power assay (FRAP), and the typical antioxidant butylated hydroxytoluene (BHT) was used as control. The results showed that the appearance,yield,chemical components and contents of the extracted essential oil by the three methods were different from each other, and the yields of the extracted oil by SD, SE and SFE-CO2 were respectively 2.3 %, 12.9 %, 7.6%; and 35, 50, 55 kind of components were respectively identified, representing 94. 64%, 99.93%, 99. 98% of the total essential oil, respectively. The extracted essential oil by SFE-CO2 scavenged DPPH radical with IC50 1.9mg/mL, and the FRAP value increased along with increasing the concentration with linear regression equation y=0. 1958x--0. 0128 (R2 = 0. 9973). The essential oil of seeds of Notopterygium incisum Ting ex H. T. Chang has some degree of antioxidant capacity, and shows the obvious dose-response relationship, but the relationship was weaker than that of synthetic antioxygen BHT. The extracted essential oil by SFE-CO2 can really,comprehensively reflect the chemical compositions of medicinal materials.
