Separation and quantitative anlysis of whey protein in south China buffalo milk by reversed-phase high-performance liquid chromatography

摘要 采用反相高效液相色谱法(RP-HPLC)对广东水牛乳乳清蛋白成分进行了分离和定量分析研究。结果表明乳清蛋白得到较好的分离且重复性良好,3种主要蛋白质(α-La、β-Lg、BSA)回归方程的线性关系>99%,且其保留时间和峰面积的RSD分别小于0.443和2.67,加标回收率范围为99.5%~101%。在相同条件下对南方水牛乳及荷斯坦牛乳进行差异性分析,表明两种乳源乳清蛋白在组分含量和遗传变异体方面均有明显的差异。 A reversed-phase high-performance liquid chromatography method for separation and quantitative analysis of China buffalo milk was proposed. The three main proteins (a-La, 13-Lg,BSA) were well separated, the retention time and the relative standard deviations (RSD) were less than 0.443 and 2.67, respectively. The linear relationship of every protein regression equation is larger than 99%. The proposed method was applied to the determination of proteins in the whey protein of buffalo milk products and the recoveries were in the range of 91%-102%. The difference of whey protein between water buffalo and Holstein milk is also studied. The result shows that two different kinds of whey proteins have significant differences in componental contents and genetic variant.
