摘要
目的观察胃癌与上皮内瘤变(GIN)的差异基因表达。方法利用基因芯片技术,筛选12例胃癌与12例GIN胃镜病理标本的差异表达基因。结果通过t检验和SAM检验筛选差异基因185个,它们与细胞黏附、血管生成、细胞增殖与凋亡等有关,与Wnt信号通路,血管内皮生长因子(VEGF)信号通路,酪氨酸蛋白激酶/信号转导及转录激活因子(JAK/STAT)信号通路,p53信号通路等细胞通路密切相关,而这些细胞通路可能与胃癌的发生有关。结论胃癌与GIN差异基因的筛选为后续胃癌分子机制的研究提供前期试验基础。
Objective To investigate the differential expression gene profile between gastric intra- epithelial neoplasia (GIN) and gastric cancer. Methods We investigated the gene expression pattern in 12 cases of gastric cancer tissues and 12 cases of GIN tissues from endoscope biopsy by gene chip. Results A group of 185 differentially expressed genes were selected by t-test and Significance Analysis of Microar- rays, and related to cell adhesion, angiogenesis, cell proliferation and apoptosis, etc. Those genes were closely related to Wnt signaling pathway, vascular endothelial growth factor (VEGF) signaling pathway, ja- nus kinase/signal transduetion and activators of transcription family(JAK/STAT) signaling pathway and p53 signaling pathway, and these pathways may contribute to the pathogenesis of gastric cancer. Conclu- sion The screening of differentially expressed genes between gastric cander and CIN may provide valuable information for further research on molecular mechanism.
