摘要
目的:深入研究中医药抗恶性肿瘤单一信号通路及多靶点联合治疗的机制。方法:经细胞培养、倒置显微镜观察细胞凋亡形态法、MTT法及流式细胞仪检测消癌解毒方及在此基础上加入激动剂脂多糖(LPS)及TLR4阻断剂(CD284)对人肝癌细胞SMMC-7721的TLRs/NF-κB信号转导通路的影响。结果:消癌解毒方具有良好的抑制人肝癌细胞SMMC-7721增殖的作用,空白、低、中、高各浓度含药血清组加入LPS后,人肝癌细胞抑制率与凋亡率均较相应浓度含药血清组下降,其中高浓度含药血清剂量组及加入CD284后对人肝癌细胞增殖的抑制作用最明显。结论:消癌解毒方的抗肿瘤机制可能是含药血清与CD284能协同作用于人肝癌细胞SMMC-7721的TLRs/NF-κB信号转导通路,抑制核转录因子NF-κB的异常持续活化,促进肿瘤细胞凋亡。
Objective: To deeply study the mechanism of single signal pathway of TCM resisting malignancy,and of multi-target combined therapy of TCM.Methods: Cell culture,inverted microscope cell apoptosis form method,MTT,and flow cytometry were used to determine the effects of Xiaoai Jiedu Fang and the ones added lipopolysaccharide(LPS) agonist and TLR4 blocker CD284 onTLRs/NF-κB signal transduction pathway of human hepatocarcinomaSMMC-7721.Results: Xiaoai Jiedu Fang had a significant effect on inhibiting the proliferation of SMMC-7721.The inhibition rate and apoptosis rate of human hepatocarcinoma were reduced after LPS agonist was added into different groups of serum containing medicine with zero,low,middle,and high concentration in turn,compared with the relevant groups of serum.The group of serum with the high medical concentration and the one added CD284 had the most significant effect on the inhibition of human hepatocarcinoma proliferation.Conclusion: The anti-tumor mechanism of the Xiaoai Jiedu Fang may be that theserum containing medicine and CD284 can promote the apoptosis of cancer cells,by working together on TLRs/NF-κB signal transduction pathway of human hepatocarcinoma SMMC-7721 which inhibits the abnormally persistent activation of NF-κB.
