摘要 目的 :探讨人肺癌组织细胞自发凋亡的发生 ,p5 3基因及凋亡相关基因bcl 2、Bax在肺癌组织中的表达及与肺癌细胞凋亡和生物学行为的关系。方法 :用甲基绿 派诺宁染色检测 5 0例肺癌组织切片中凋亡细胞 ,LSAB免疫组化法检测 p5 3和Bax的蛋白表达 ,原位杂交方法检测bcl 2mRNA的表达。结果 :10 0 %小细胞肺癌凋亡发生数 10个 /HPF ,2 1%的肺鳞癌和 15 %的肺腺癌凋亡发生数 >10个 /HPF。p5 3与bcl 2的表达与肺癌淋巴结转移之间有显著统计意义 (P <0 0 5 ) ,p5 3的表达与bcl 2和Bax表达之间无统计学显著意义。结论 :小细胞肺癌较肺鳞癌和肺腺癌更易于发生自发凋亡 ,肺癌组织中 p5 3表达的改变并不一定伴有bcl Purpose To better understand the apoptosis, the expression of p53, bcl 2 and Bax in human lung carcinoma and the relationship between the expression and the biological behavior. Methods Fifty cases of human primary lung carcinomas were selected. The apoptosis, the protein expression of p53, Bax, and the mRNA expression of bcl 2 were detected using Methyl Green Pyronine staining, immunohistochemistry,and in situ hybridization. Results The apoptosis numbers were more than 10/HPF in all 5 cases SCLC, only in 21% squamous cell carcinoma and 15% adenocarcinoma. There was significantly statistic difference between the expression of p53 protein and bcl 2 and the metastatic status of lymph node( P <0 05),but no statistic difference between expressions of p53 protein, bcl 2 and Bax. Conclusion Apoptosis happens much easier in small cell lung cancer than in squamous cell carcinoma and adenocarcinoma. p53 protein overexpressions do not accompany changes in bcl 2 and Bax overexpression.
