摘要
目的研究SB-431542对树突状细胞(DC)融合疫苗的制备和抗小鼠胰腺癌效应的影响。方法(1)SB-431542(转化生长因子-β1受体阻滞剂)与粒细胞集落刺激因子、白细胞介素4、脂多糖联合应用,刺激C57BL/6小鼠骨髓来源DC成熟,应用聚乙二醇融合技术融合DC和小鼠胰腺癌细胞,制备融合疫苗。②观察细胞形态、细胞表面表达、MTT比色法检测检测肿瘤细胞体外杀伤细胞毒活性。③制备荷瘤胰腺癌小鼠模型,分三组分别观察肿瘤体积大小,生存期观察。结果①在SB-431542参与下,成熟DC的细胞数目及形态无显著改变。②DC表面CD80、CD86、CD40的表达较未加SB-431542显著增高;MTT比色法,添加SB-431542的DC疫苗组对肿瘤细胞的杀伤抑制率显著提高。③体内抗肿瘤效应SB-431542的DC疫苗组肿瘤大小明显减小.生存期显著提高(P〈0.05)。结论SB-431542组融合细胞的分子表达较非SB-431542组显著提高,体外体内均显示SB-431542组有更好的抗肿瘤效应,说明SB-431542对增强融合疫苗抗胰腺癌特异性免疫效应有明显的作用。
Objective To investigate the effect of SB-431542 on the preparation of the fusional vaccine with dendritic cells and pancreatic cancer cells, and the anti-tumor immunological reaction of the vaccine. Methods (1)In the presence of SB-431542 ( an inhibitor TGF-β1 receptor) GM-CSF, IL-4 and LPS, the maturation of dendritic ceils was made from C57BL/6 mice bone marrow. The preparation of the fusional vaccine was made in the presence of PEG. (2)The biologicalcharacteristics of fusion cells, including their morphological specialties, the expression of cell surface phenotypes, inducing specific cytotoxic T lymphocyte (CTL). (3)The survical period and the tumor regression were observed on murine pancreatic carcinoma. Results (1)SB-431542 can stimulate the full maturation of dendritic cells. Dendritic cells and paneO2 cells could be fused by PEG. (2)The fusion cells exhibited typical biological characteristics. The expression of cell surface phenotypes enhance efficiently. (3)The tumor size was obviously regressed and the survival period was obviously prolonged in the treatment group than that in other groups (P 〈 0.05). Conclusions SB-431542 can enhance the maturation of dendritic cell. The fusion vaccine could induce a specific and effective immune response and chemotherapeutic function against tumor cell implanted in mice.
