摘要
目的建立测定贯叶连翘提取物中金丝桃苷含量的高效液相色谱法。方法采用Agilent ZORBAX SB-C18(250 mm×4.6 mm,5μm)色谱柱,以乙腈-0.1%磷酸溶液(15∶85)为流动相,流速1.2 ml/min,检测波长256 nm;柱温为35℃。结果金丝桃苷检测浓度在11.4~91.2μg/ml范围内与峰面积积分值呈良好的线性关系(r=0.999 9);平均回收率为100.7%,RSD为1.37%。结论该法快速、简便、准确,重复性好,可用于贯叶连翘提取物中金丝桃苷的含量测定。
Objective To develop an RP - HPLC method for the determination ot hypenn m ttypertcum perJoratum L. extracts. Methods The RP -HPLC system consisting of Agilent ZORBAX SB -C18 (250 mmx 4.6 mm, 5 μm) column was adopted and the mobile phase was acetonitrile-0.1% phosophoric acid( 15: 85) at the flow rate of 1.2 ml/min; the detection wavelength was 256 nm and the column temperature was 35℃. Results The linear range for hyperin was 11.4 - 91.2 p,g/ml ( r =0. 999 9). The average recovery was 100.7% (RSD = t. 37% ). Conclusion This method is simple,rapid,accurate,and it can be used for the quality control of the Hypericum pe(foratum L. extract.
