松穴褥盘孢菌生物学特性的研究 被引量:12

摘要 松穴褥盘抱菌(Dothistroma pini Hulbary)在PDA固体培养基上生长缓慢,7d后可产生大量分生孢子,并产生红色素。在麦芽汁液体培养基上生长好。菌丝在液体培养基上生长最适温度为15—30℃,在20℃时产孢量最大。麦芽糖为菌丝生长和产孢最佳碳源。色氨酸为菌丝生长最佳氮源,而酒石酸胺和天门冬素为产孢最佳氮源。菌丝生长最佳pH值为3—5,最佳产孢pH值为5。维生素对菌丝生长无显著影响,维生素H能促进产孢。微量元素对菌丝生长也无显著影响,但铁能促进产抱。分生孢子在2%麦芽汁中萌发率最高。相对湿度低于98%时不葫发。分生孢子萌发最适温度为15—20℃。不同光照对孢子萌发影响不大。 The study was made on the biological nature of Dothistroma pini Hulbary. It grows slowly on PDA solid culture medium and may produce a lot of spores and red pigment. But, it grows fast in liquid culture medium of malt agar. The optimum temperature for the mycelium growing in liquid culture medium is 15—30℃, and spores production is the most at the temperature of 20℃. Maltose is the best carbon source for spores production and mycelium growth. Trytophan is the best nitrogen source for mycelium growth and asparagine and ammonium fartrade for spores production. pH 3—6 is the most suitable for mycelium growth and pH 5 for spores producrion. Vitamin and trace elements have no remarkable effect on mycelium growth but vitamin H and trace element Fe can promote spores production. Spores germinate best in 2% malt extract and the optimum temperature for germination is 15—20℃. Spores can not germinate when relative humidity is bellow 98%. Illumination makes no difference for spores germination.
