Apollon siRNA抑制肺癌细胞生长并提高放疗敏感性 被引量:3

摘要 目的调查与Apollon小干扰RNA(siRNA)并用是否可以提高人肺癌细胞放疗敏感性。方法采用转染野生型及突变型p53基因的两种非小细胞肺癌细胞株H1299/wtp53及H1299/mp53,通过siRNA技术沉默癌细胞Apollon表达,并以实时定量聚合酶链反应(RT-PCR)检测Apollon基因表达。采用细胞克隆生成分析确定Apollon沉默的抗癌效果及放疗增敏作用。结果以Apollon为靶点的siRNA转染细胞后,可观察到Apollon基因表达抑制。Apollon siRNA可抑制癌细胞增殖并提高H1299细胞放疗敏感性。结论在肺癌放疗中,Apollon siRNA可能是一个潜在的放疗增敏剂候选者。
