摘要
目的观察蜈蚣三七总皂苷对胶原性关节炎(CIA)小鼠炎症细胞因子的影响,探讨其免疫作用机制。方法建立Ⅱ型胶原诱导的CIA小鼠模型,随机分为蜈蚣三七总皂苷组(104 mg/kg)、阳性对照组(雷公藤甲素66μg/kg)、模型组、正常组,致炎7 d后各组均开始灌胃给药,连续4周,光镜下观察各组动物足趾关节的病理变化,ELISA法检测CIA小鼠血清中白介素(IL)-2、IL-4、IL-6、IL-10、IL-12及γ-干扰素(IFN-γ)的水平。结果蜈蚣三七总皂苷可明显减轻关节软骨细胞和滑膜增生,抑制炎性细胞浸润。与模型组比较,蜈蚣三七总皂苷可明显提高IL-4、IL-10及IL-12的分泌水平(P<0.01),降低IL-6和IFN-γ的分泌水平(P<0.01),对IL-2的表达无明显影响。结论蜈蚣三七总皂苷通过调节细胞因子含量,纠正Th1/Th2的失衡状态,在防治CIA中发挥免疫调节的作用。
Objective To investigate the effects of total saponins from Anemoneflaccida (TSA) on the levels of cy- tokines in mice with collagen II-induced arthritis (CIA). Methods CIA model in mice were obtained by the in- fection of collagen 11. Then mice were randomly allocated into four groups : TSA ( 104 mg/kg) , Tripterygium wil- fordii (as positive control, 66μg/kg) , model control and normal control. After four weeks of oral administration, the arthral pathology of all mice were observed by optical microscope, the levels of cytokines in serum were detected by ELISA. Results TSA could significantly improve the pathological histology in cartilage and synovium, and sup- press the inflammatory cells infiltration in joints of CIA mice. The levels of interleukin (IL) -4, IL-10 and IL-12 in- creased markedly and those of IL-6 and interferon (IFN) -γdecreased significantly in TSA group when compared to model control (P 〈 0. 01 ). Conclusion TSA may modulate the levels of cytokines and correct the imbalance state of Thl and Th2 in CIA. TSA has immune regulative effects as a treatment for CIA.
