黄芪杂多糖通过调节IL-6和PG-1的表达提高仔猪免疫力 被引量:2

摘要 为探明AHPS对仔猪的免疫调控作用,通过细胞的分离培养技术、MTT法、ELISA检测和实时荧光定量PCR技术对比不同剂量黄芪杂多糖(AHPS)注射组与空白对照组的仔猪血液内的T淋巴细胞和单核细胞的增殖活性、IL-6含量,骨髓中抗菌肽Protegrin-1(PG-1)基因的表达水平,并进行相关性分析。结果显示,AHPS对仔猪外周血中的T淋巴细胞和单核细胞的增殖有显著的促进作用;IL-6含量随着注射时间的延长持续增多,在同一时间内呈剂量依赖型增加;AHPS能够显著上调骨髓中PG-1mRNA的表达且呈剂量依赖性增加。结果表明,AHPS可能通过促进仔猪外周血中T淋巴细胞和单核细胞的增殖来提高血清中IL-6的含量,并诱导PG-1mRNA的表达,从而提高仔猪机体免疫力。 To study the immunoregulatory effect of Astragalus heteropolysaccharides (AHPS) on piglets, the proliferation activity of T lymphocytes and monocytes in peripheral blood were observed by MTT, the IL-6 contents in serum were determined by ELISA, the expression level of PG-1 mRNA in marrow was evaluated with RT-qPCR. The results were as follows: AHPS could promote significantly proliferation activity of T lymphoeytes and monocytes in peripheral blood; IL-6 contents in serum increased significantly with the extension of the injection time, and dose-dependent patterns at the same time; AHPS could increase significantly expression of PG-1 mRNA and dose-dependent patterns. The results suggested that AHPS may improve the immunity of piglets by increasing IL-6 contents and inducing the expression level of PG-1.
