摘要
目的:观察胆总管短暂梗阻(TCBDO)对小鼠肾脏缺血再灌注(KIR)损伤的影响。方法:小鼠30只,采用胆总管结扎术建立TCBDO模型,于术后第0、1、2、3、4及5天测血清总胆红素(TB)、丙氨酸氨基转移酶(ALT)、肌酐(Cr)水平,于术后第3、4及5天测肾脏HSP27蛋白的表达。另取小鼠40只,随机分为4组,每组10只,分别给予胆总管结扎假手术、胆总管结扎术、胆总管结扎假手术+KIR造模术和胆总管结扎+KIR造模术,胆总管结扎术后第4天做KIR;缺血25min再灌注24h后,留取血样测血清Cr水平;留肾标本做PAS染色,观察形态学改变,MPO免疫组化染色分析肾中性粒细胞浸润情况,TUNEL法检测肾脏凋亡细胞。结果:TCBDO后小鼠血清TB和ALT水平快速上升,后迅速降低(P<0.001);血清Cr水平变化不大(P>0.05);肾脏HSP27蛋白表达水平增高(F=153.520,P<0.001)。与胆总管结扎假手术+KIR造模术组比较,胆总管结扎+KIR造模术组小鼠血清Cr水平(F交互=72.405,P<0.001)、肾组织病理评分(P<0.05)、中性粒细胞浸润程度(P<0.05)和凋亡细胞数(F交互=29.338,P<0.001)均降低。结论:TCBDO可能通过促进小鼠肾脏HSP27的表达,从而减轻肾KIR损伤。
Aim:To investigate the effects of transient common bile duct obstruction(TCBDO) on expression of heat shock protein 27(HSP27) and ischemia-reperfusion in kidneys(KIR) of mice.Methods:A total of 30 mice were performed common bile duct ligation(CBDL) to build TCBDO model, and the serum total bilirubin(TB), alanine aminotransferase(ALT),and creatinine(Cr) were measured at day 0,1,2,3,4 and 5 after CBDL, the expression of HSP27 protein in kidney tissue was detected at day 3,4,and 5 after CBDL. A total of 40 mice were divided into 4 groups randomly (10 in each group) and underwent CBDL sham surgery only,CBDL, CBDL sham surgery combined with KIR,and CBDL combined with KIR,respectively. KIR surgery was performed 4 days after CBDL.The serum Cr level was determined,morphological changes and neutrophil infiltration of the kidneys were observed by PAS and MPO immunohistochemical staining,respectively and cell apoptosis was detected by TUNEL.Results: After CBDL, the serum TB,ALT levels,but not Cr,changed significantly, and HSP27 protein expression in kidney tissue upregulated(F=153.520,P0.001).In CBDL combined with KIR group,the serum Cr level decreased(F=72.405,P0.001),and renal tissue injury score(P0.05), neutrophil infiltration(P0.05) and apoptosis(F=29.338,P0.001) induced by KIR mitigated.Conclusion: TCBDO could attenuate KIR injury in mice probably through promoting the expression of HSP27 protein.
