摘要
目的:研究菊明提取物中降压活性物质黄酮类成分的质量控制标准。方法:以蒙花苷和野菊花对照药材为对照品,采用薄层色谱法(TLC)鉴别方中野菊花。以芦丁为对照品,采用亚硝酸钠-硝酸铝-氢氧化钠显色法测定总黄酮含量;采用高效液相色谱法(HPLC)测定蒙花苷含量,Discovery C18(250mm×4.6mm,5μm),流动相为甲醇-2%醋酸(52:48),检测波长为334nm。结果:薄层斑点清晰,专属性强。芦丁对照品线性范围在13.37-66.84μg/mL,回归方程为Y=0.0129X-0.0116,r=0.9999;平均回收率为98.28%,RSD为2.05%。蒙花苷对照品线性范围在21.90-1 752.00μg/mL,回归方程为Y=92845X+401863,r=0.9999;平均回收率为101.83%,RSD为1.10%。结论:建立的薄层鉴别和含量测定方法简便可行、结果准确,可用于菊明提取物中黄酮类成分的质量控制。
Objective: To study the quality control standard of flavonoids from Juming extracts. Methods: TLC was adopted to distinguish the Chrysanthemi Indici Flos with rutin and the medicinal material as reference substance. The content of total flavonoids was determined by NaNO2-Al(NO3)3-NaOH with rutin as reference substance. Linarin was determined by HPLC with C18 (250mm×4.6mm, 51μm), methanol-2%HAc (52:48) as mobile phase and 334nm as detection wavelength. Results: The spots in TLC were clear and distinctive. Linear relation of rutin was 13.37-66.84μg/mL and the regression equation was Y=0.0129X-0.0116, r=0.9999. The average recovery was 98.28% with RSD 2.05%. Linear range of linarin was 21.9-1 752μg/mL and the regression equation was Y=92845X+401863, r=0.9999. The average recovery rate was 101.83% with RSD 1.10%. Conclusion: The methods were simple and accurate. It can be used for the quality control standard of Juming extracts.
