摘要
目的找出乙二胺四乙酸二钾(EDTA-K2)与SD大鼠全血比例的最佳范围,观察不同浓度EDTA-K2处理的SD大鼠全血标本长时间冰箱保存对血细胞指标的影响。方法连续4 d用XT-2000i型全自动血细胞分析仪对经4个不同浓度EDTA-K2处理的10只SD大鼠全血标本(共40个)进行检测,其测定结果用SPSS17.0做统计学分析。结果采血0 h时,10 mg/ml组中性细胞百分率比4 mg/ml组降低(P<0.05)。与0 h比较,24 h后血小板计数(PLT)、平均血小板体积(MPV)、红细胞平均体积(MCV)即有明显变化,差异有统计学意义(P<0.05);48 h后血红蛋白(HGB)、白细胞计数(WBC)、中性细胞绝对值(NEUT#)也出现明显变化,差异有统计学意义(P<0.05);72 h后红细胞计数(RBC)、中性细胞百分率(NEUT%)、淋巴细胞百分率(LYMPH%)均出现明显变化,差异有统计学意义(P<0.05)。结论 EDTA-K2与SD大鼠血液比例为4.0~8.0 mg/ml时,血标本效果较为理想。在4℃保存条件下SD大鼠血标本也不宜放置24 h以上,应及时检测,以提高血细胞各种参数准确性。
Objective To find the best range of the ethylenediamine tetraacetic acid dipotassium ( EDTA - K2 ) and SD rat complete blood, and observe the influence on blood cell index caused by long - time storage of SD rat complete blood sample treated by different concentration of EDTA - K2. Methods Detecting 40 SD rat complete blood samples (10 rats) which were treated by different concentration of EDTA -K2 by using XT -2000i automatic blood cells analyzer for 4 days. The results are subjected to statistic analysis with SPSS17.0. Resuits : at 24 h, percentage of 10mg/ml neutral ceils is significantly reduced ( P 〈 0. 05 ) compared with that of 4 mg/ml. PLT, MPV, MCV at 0 h are significantly different from those at Oh. HGB, WBC, NEUT# and RBC after 48 h as well as NEUT% and LYMPH% after 72 h also have significantly difference. Conclusion When the ratio of the EDTA - K2 to the SD rat blood is ranging from 4. 0 to 8.0 mg/ml, the blood samples are ideal. The SD rat blood samples are not suitable for being stored for over 24 h at 4 DEG C and are required to be detected in time, so that the accuracy of various parameters of the blood cells can be improved.
