摘要
拟合成并鉴定林可霉素(Lincomycin,LIN)人工抗原,通过动物免疫生产亲和力高、特异性好的鼠源LIN多克隆抗血清。采用高碘酸钠法将林可霉素与牛血清白蛋白(BSA)及鸡卵清蛋白(OVA)分别偶联,合成免疫原LIN-BSA和包被原LIN-OVA,并采用紫外分光光度法(UV)和聚丙烯酰胺凝胶电泳(SDS-PAGE)进行鉴定。结果表明,半抗原LIN和载体蛋白偶联成功。动物免疫试验(免疫BALB/c纯系小鼠)结果显示,3只小鼠效价均达1∶3 000,且2号小鼠多抗血清抗LIN的半数抑制质量浓度IC50为3.85μg/L,与莫能菌素的交叉反应率为6.19%,与其他药物无交叉反应。表明成功获得高效价、特异性好、亲和力高的鼠源抗LIN多抗血清。
The goal of this study was to synthesize artificial antigen of lincomycin and to obtain its mice polyclonal antiserum.Artificial antigen LIN-BSA and LIN-OVA were synthesized using sodium periodate by linking carrier proteins BSA and OVA to LIN.The antigen LIN-BSA and LIN-OVA was identified by ultraviolet scanning and SDS-PAGE.The results showed that the hapten LIN was successfully linked to carrier proteins.The animal immunization results showed that three BALB/c mice indirect ELISA titer against LIN were 1∶3 000 and the IC50 of No.2 mice was 3.85 μg/L and the high-titer;the rate of cross reaction of LIN mAb with Monensin was 6.19%,and there was no cross-reactivity to other compounds;sensitivity LIN polyclonal antiserum has been generated.The artificial antigen of LIN and its mice polyclonal antiserum laid a foundation base for product the monoclonal antibodies and rapid test reagent against LIN.
