摘要
目的:研究加味二至丸对血管性痴呆的治疗作用,并探索其作用机制。方法:用双侧颈内动脉结扎(2-VO)法制作大鼠脑缺血再灌注血管性痴呆(VD)模型(女贞子、墨旱莲、丹参、天麻)和肾上腺素合2-VO法制作大鼠气滞血瘀证VD模型,分别以2.66、1.33、0.67g/kg灌胃模型大鼠分别30天,以跳台实验评价学习记忆能力,血液流变仪测定血液流变学参数,酶联免疫法测定大脑海马Ach、AchE、大脑皮层5-HT、NE、DA含量及血清ET、TXB2、6-Keto-PGF1α含量;毛细管电泳法测定海马氨基酸Glu、Gly、Asp、GABA含量。结果:给予加味二至丸2.66g/kg能明显缩短脑缺血再灌注血管性痴呆大鼠跳台试验的潜伏期和减少其5min内错误次数;2.66、1.33、0.67g/kg均能明显减少气滞血瘀证脑缺血再灌注血管性痴呆大鼠5min内错误次数,且对该模型潜伏期有延长趋势。加味二至丸能显著增加海马中Ach含量而降低AchE活性,显著增加大脑皮层DA含量,对5-HT、NE含量亦有增加趋势;能调节痴呆大鼠海马氨基酸GABA、Glu、Gly、Asp的含量;同时可明显改善模型大鼠的血液流变学参数,显著降低血清TXB2、ET水平而升高6-Keto-PGF1α、CGRP水平。结论:加味二至丸具有治疗血管性痴呆作用,其机理可能与海马Ach含量和降低AchE活性、调经大脑神经递质、调节血管活性物质、改善血液流变性有关。
Objective: To validate the curative effect of modified Erzhi Pill to vascular dementia by zoopery, at the same time try to understand the pathogenesis of vascular dementia. Methods: Adopting bilateral cervical arteries ligation on rats to make the animal model of cerebral ische- mia and reperfusion; as for the other one animal model, giving epinephrine before surgery; respectively filling rats'stomach with liquid medi- cine of Modified ErZhi Pill as 2.66,1.33,0.67g/kg for a month. Then detecting the purity of Ach, AchE, GABA, Glu, Gly, Asp in hippo-campus; 5-HT, NE, DA in pallium and ET, TXB2 , 6-Keto-PGFIot, CGRP in serum of rats. Results: When the modified Erzhi Pill was giv- en cerebral ischemia and reperfusion vascular dementia rats as 2.66g/kg, it could obviously shorten the latentperiod and reduce the errorcou- nts in platform experiment; As for cerebral ischemia and reperfusion vascular dementia rats of Qi stagnancy and blood stasis syndrome ,the er- rorcounts also could be clearly decreased when the modified Erzhi Pill~ dosage is on 2.66, 1.33 or 0.67g/kg. Modified Erzhi Pill have in- creased the purity of Ach, GABA, Glu, Gly, Asp in hippocampus and 5-HT, NE, DA in pallium and lowered the activity of AchE ; besides, the Pills had significantly lowered ET, TXB2,6-Keto-PGF1 et, CGRP in serum of rats. Conclusion: Modified Erzhi Pill could preferably play the role to treat dementia on account of its effect. The mechanism is probabely relevant to the purity of Ach in hippocampus, lowering the activity of AchE; to adjust the brain neurotransmitters and vascular active material; to improve blood rheology and so on.
