Synthesis of Trimetazidine Hydrochloride

摘要 研究一种冠状动脉血管扩张剂盐酸曲美他嗪的合成方法。以六水哌嗪为起始原料,经酰化、Mannich反应、水解、成盐反应制得目标化合物。目标化合物的结构经熔点测定、红外及1 H-NMR谱确证,总收率为29.2%,经HPLC检测纯度为98.5%。该合成方法成本较低、操作简便、条件温和,适于工业生产。 The synthetic method of trimetazidine hydrochloride, a coronary vasodilator agent, was studied with piperazine hexahydrate as the starting material. The target compound was obtained by the acylation of the raw materials, mannich, hydrolysis, and saltforming with hydrochloric acid. Its structure was identified by melting points, IR and 1H-NMR techniques. The overall yield was 29.2% and the purity (HPLC) was 98.5%. This method of synthesis can be used in- dustrially for its low cost, simple operation, and mild reaction conditions.
