穿山龙总皂苷对CIA大鼠滑膜血管内皮生长因子及其受体Flk-1的影响 被引量:9

摘要 目的:研究穿山龙总皂苷(TSRDN)对CIA大鼠滑膜血管新生的影响及其作用机制。方法:建立CIA大鼠模型,造模后给予TSRDN、雷公藤和薯蓣皂苷元灌胃,并采用免疫组化检测滑膜组织MVD、VEGF及Flk-1表达。结果:CIA模型组大鼠滑膜新生血管增多,MVD、VEGF及Flk-1表达升高(P<0.01),给药后大鼠滑膜新生血管数量减少,MVD、VEGF和Flk-1表达降低(P<0.01)。结论:穿山龙总皂苷可能通过抑制VEGF和Flk-1的表达来发挥抗血管新生作用。 Objective:To study effects of total saponin of Rhizoma Dioscreae Nipponicae(TSRDN) on angiogenesis in synovial tissue of CIA rats,and investigate the mechanism of TSRDN on antiangiogenesis in treating rheumatoid arthritis.Methed: After the CIA rat model was established,the rats in treatment group were lavaged with TSRDN,tripterygium,Diosgenin.Immunohistochemical staining was used to observe MVD,VEGF and Flk-1.Results: MDV,VEGF and Flk-1 expression of CIA model group were obviously higher than normal group(P0.01).After treatment,MDV,VEGF and Flk-1 expression were obviously lower than CIA model group(P0.01).Conclusions: TSRDN can inhibit angiogenesis in synovial tissue by down regulating VEGF and Flk-1 expression.
