摘要
目的:观察淫羊藿总黄酮对酗酒模型小鼠精子质量的影响。方法:将50只小鼠随机分为正常对照组、模型组和TFE高、中、低3个剂量组,除正常组动物灌胃等体积生理盐水外,其余4组动物均按10 mL.kg-1体质量灌胃给予20%乙醇溶液,连续造模30 d。于第15天开始,各组分别灌胃给予药物或生理盐水,连续用药15 d。末次给药后2 h,处死小鼠,制备附睾精子悬液,分别测定睾丸/附睾质量指数、精子数量、精子活力。采用单细胞电泳法测定精子DNA损伤。结果:TFE能显著地提高酗酒小鼠的睾丸/附睾质量指数和精子数量,增加精子的活力和降低精子的畸变率,对酒精造成的DNA损伤具有较好的拮抗作用。结论:淫羊藿总黄酮对酗酒小鼠的生精功能有一定的改善效果。
OBJECTIVE To observe the effects of total flavonoids of Herba epimedii（TFE） on the sperm quality of intemperance model in mice.METHODS Fifty Kunming mice were divided into 5 groups at random.The mice were treated with 20% ethanol solution（10mL/kg） through gastric perfusion in four groups for 30 days respectively except the normal control group which treated with the same volume of normal saline（NS）.From the 15th day,the mice were given i.g.with drugs or NS successively.2h later after last administration,the mice were killed and the sperm suspension was made.Testis/epididymis weight index,sperm quantity,sperm vitality were determined.Single cell gel electrophoresis（SCGE） was used to determine the damage of sperm DNA.RESULTS The results showed that the TFE could significantly improve thedecrement of testis weight index testis（TWI）/ epididymis weight index（EWI）,sperm quantity and vitality and reduce sperm aberration rate induced by alcohol.It could also antagonism the damage of sperm DNA by alcohol.CONCLUSION TFE can significantly improve the spermatogenic function of intemperate mice.
