D-101树脂纯化胭脂虫红色素的工艺研究 被引量:1

摘要 为了提高胭脂虫红色素的纯度,研究了D-101树脂纯化胭脂虫红色素的工艺。比较了不同pH值预处理胭脂虫红色素提取液的吸附效果,研究了树脂动态吸附和洗脱动力学曲线,并对树脂吸附条件和洗脱条件进行了研究。优化得到D-101树脂纯化胭脂虫红色素的最佳工艺条件为:提取液采用pH为3进行预处理,上柱液体积为2BV,上柱流速为3BV/h,体积分数为75%的乙醇洗脱,洗脱液体积为3.5BV,洗脱速率为2BV/h,在此条件下,吸附量为42.15mg/mL树脂,洗脱率高达98.24%,胭脂虫产品纯度可达58.47%,能够满足一般食品工业的对胭脂虫红色素的纯度要求。采用D-101大孔吸附树脂分离提纯胭脂虫红色素具有吸附量较大、洗脱率高、产品纯度较高、适合工业化大规模生产等优点,具有广阔的工业应用前景。 In order to improve the purification of cochineal pigment, the purification technology by D-101 resin was studied. The adsorption effects of different pH value pretreating cochineal pigment extracts were compared. The dynamic adsorption curve and dynamic elution curve were studied. And the dynamic adsorption and desorption parameters for the purification cochineal pigment by D-101 resin were investigated. Results showed that the optimal purification conditions of cochineal pigment by D-101 resin were found to be: the pretreatment pH value of cochineal pigment extracts was 3, the adsorption volume of cochineal pigment was 2 BV, adsorption rate was 3 BV/h, 75% volume percent ethanol was used as elution solvent, the volume of elution solvent was 3.5 BV, the elution rate was 2 BV/h. Under these optimum conditions, the adsorption volume was 42.15 mg/mL resin, the elution rate was as high as 98.24%, the purity of the final products was 58.47%, and the purity could reach the demand of general food industry for cochineal pigment. This technology for purifying cochineal pigment had a lots of good advantages, such as biggish adsorption volume, high elution, high product purification, and suitable for industrial production. So it had wide application prospect in industry.
