摘要
为探讨不同培养条件对白花丹参组培苗玻璃化的影响,以白花丹参不育变异品种的幼嫩茎段为外植体,以MS为基本培养基,研究了KT/NAA比例、活性炭添加及琼脂浓度对防治白花丹参组培苗玻璃化现象的作用。结果表明,当KT浓度为1.5 mg.L-1、NAA浓度为0.5 mg.L-1、培养基中添加0.3%活性炭、琼脂浓度为0.8%时,白花丹参组培苗增殖系数较高,玻璃化率低,炼苗成功率最高。因此,MS+KT 1.5 mg.L-1+NAA 0.5 mg.L-1+琼脂0.8%+活性炭0.3%的培养基可以有效防治白花丹参组织培养中的玻璃化现象。
This paper addresses the impacts of different culture conditions on the vitrification of Salvia miltiorrhiza Bge.f.alba seedlings.We investigated the influences of KT/NAA ratio,active carbon and agar concentration on the prevention of vitrification of tissue culture seedlings with the tender stems of Salvia miltiorrhiza Bge.f.alba C.Y.Wu et H.W.Li.‘buyu’ as the explants and MS as basic medium.Experimental results show that the tissue culture seedlings has higher bud multiplication coefficient and lower vitrification rate when KT concentration of 1.5 mg·L-1,NAA concentration of 0.5 mg·L-1,active carbon of 0.3% and agar concentration of 0.8% are employed.Therefore,the MS+KT 1.5 mg·L-1 +NAA 0.5 mg·L-1 + agar 0.8% +active carbon 0.3% medium can effectively prevent vitrification of plantlets.
