Simultaneous determination of huperzine A and huperzine B in different parts of Huperzia serrata from different habitats by HPLC

摘要 目的:测定不同产地、不同部位千层塔药材中石杉碱甲和石杉碱乙的含量。方法:采用UItimate XB-C18(4.6 mm×250 mm,5μm)色谱柱,流动相为乙腈-甲醇-水(10∶55∶240),含0.32%磷酸和0.5%三乙胺,流速1.0 mL.min-1,检测波长309 nm,柱温25℃。结果:测得各地千层塔中石杉碱甲、石杉碱乙最高含量分别为0.0900%和0.0183%,各地石杉碱甲、石杉碱乙最低含量分别为0.0014%和0。结论:所测定的方法简便、可靠、快速,重复性好,结果稳定,适用于千层塔药材中石杉碱甲和石杉碱乙的同时快速含量测定,该研究可为千层塔最佳产地及最佳药用部位的确定提供参考。 Objective: To develop an HPLC method for simultaneous determination of huperzine A ( Hup - A) and huperzine B (Hup -B) in different parts of Huperzia serrate from different habitats. Methods:The assay was per- formed on an Uhimate XB -C18(4.6 mm ×250mm,5 μm) column with the mobile phase of aeetonitrile- metha- nol - water ( 10 : 55 : 240 ) containing 0. 32 % phosphoric acid and 0.5 % triethylamine. The flow rate was 1.0 mL .min-1 ,the detection wavelength was set at 309 nm, and the column temperature was set at 25 ℃. Re- sults:The contents of huperzine A and huperzine B in Huperzia serrata collected at different habitats were evidently different. The maximum contents of huperzine A and huperzine B were 0. 0900% and 0. 0183% , respectively;the minimum contents of huperzine A and huperzine B were 0. 0014% and 0,respectively. Conclusions:The method is simple, accurate and repeatable, which provides a reliable way for evaluating the quality of Huperzia serrata by meas- uring huperzine A and huperzine B, as well as provide a reference for the determination of the best origin and the most rational part of the plant.
