摘要
目的:研究鲜壁虎水提物与醇提物体内、外的抗肿瘤活性。方法:体外采取MTT法测定鲜壁虎水提物、醇提物对BEL-7402细胞和HepG2细胞生长的抑制作用。体内复制移植瘤小鼠H22模型,给药后计算抑瘤率、胸腺指数和脾脏指数。结果:0.0625mg·mL-1壁虎水提物对HepG2细胞生长有显著抑制作用(P<0.05),1.0000、0.2500、0.0625mg·mL-1壁虎水提物对BEL-7402细胞的生长均有显著抑制作用(P<0.05)。1.0000、0.2500、0.0625mg·mL-1壁虎醇提物对2种细胞有促进增殖的趋势。1.0000、0.2500、0.0625mg·mL-1壁虎水提物与1.0000、0.0625mg·mL-1壁虎醇提物均可显著抑制H22荷瘤小鼠肿瘤生长活性(P<0.05)。结论:鲜壁虎水提物体外有抑制肿瘤细胞生长的作用,醇提物有促进细胞增殖的趋势。鲜壁虎水提物、醇提物体内均有抗肿瘤活性,水提物抑瘤率高于醇提物。
OBJECTIVE: To study the in vitro and in vivo antitumor activity of the aqueous and ethanol extracts from Gekko swinhonis Gunther. METHODS: The inhibitory effects of aqueous and ethanol extracts from G. swinhonis on proliferation of BEL-7402 cell and Hep G2 cell in vitro were measured by MTT assay. The in vivo H22 tumor mice model was induced to calculate anti-tumor rate, thymus gland index and spleen index. RESULTS: The aqueous extracts of G. swinhonis Guther 0.062 5 mg. mL^-1 inhibited the proliferation of Hep G2 cells. The aqueous extracts of G. swinhonis Guther 1.000 0,0.250 0,0.062 5 mg.mL^-1 inhibit- ed the proliferation of BEL-7402 cells. The water extracts from G. swinhonis Guther 1.000 0,0.250 0,0.062 5 mg.mL^-1 and the eth- anol extracts from G. swinhonis Guther 1.000 0,0.062 5 mg. mL^- 1 promoted the proliferation of 2 kinds of cells. The ethanol extracts from G. swinhonis Guther 1.000 0,0.250 0,0.062 5 mg. mL^-1 inhibited the activity of tumor in H22 hepatocarcinoma-bearing mice. CONCLUSION: The agueeus extracts of G. swinhonis Guther can inhibit the growth of tumor cell in vitro, while ethanol ex- tract of it promote the proliferation of tumor cell. They all have antitumor effect, but the in vivo antitumor effects of aqueous ex- tracts of G. swinhonis Guther is more effective than ethanol extracts.
