摘要
目的 观察导入 baxα基因并稳定表达的人低分化鼻咽癌细胞株 (CNE- 2 B)生物学行为变化。方法 琼脂糖凝胶电泳分析 DNA片段化 ;γ-射线照射。结果 CNE- 2 B在进行正常及受 γ-射线照射后传代培养时 ,其细胞总数的增加速度比导入载体的对照组 CNE- 2 P细胞株明显减慢 ;自发性细胞凋亡数明显增加。 CNE- 2 B细胞接种到 10只裸鼠双侧背部皮下均未见形成移植瘤 (0 / 2 0 ) ,而对照的 10只裸鼠的成瘤率为 10 0 % (2 0 / 2 0 )。结论 转导人 baxα基因的CNE- 2 B细胞株的生物学行为发生了显著性改变 (P<0 .0 1)
Objective To observe the changing of biological behavior in human poorly differentiated nasopharyngeal cell line (CNE 2B), which is integrated with human bax α gene. Methods DNA fragmentation analysis by agarose gel electrophoresis; γ rays irradiation. Results CNE 2B cell line grew much slower than CNE 2P cell line (containg pSFFV neo without human baxα gene) Whether 60 Co γ ray irradiation was given or not. Spontaneous apoptotic cell number of CNE 2B increased greatly. No transplanted tumor occurred after CNE 2B cells were inoculated subcutaneously into nude mice(0/20), while CNE 2P cells' transplantability was 100 % (20/20). Conclusion The biological behavior of CNE 2B was significantly changed ( P <0 01) in compare with CNE 2P cell line as control.$$$$
