摘要
目的探索慢性阻塞性肺疾病(chronic obstructive pulmonary disease,COPD)肺泡上皮结构改变的潜在机制.方法建立大鼠COPD模型,于建模第28天,42 d分批处死大鼠,采用免疫组化观察大鼠肺泡上皮紧密连接中闭锁蛋白(Occludin)和闭锁小带蛋白1(zonula occludens-1,ZO-1)的分布和表达.结果 COPD时,42 d实验组肺泡上皮细胞Occludin和Zo-1表达明显下降(P<0.05);28 d实验组肺泡上皮细胞Occludin和Zo-1表达有所降低,但差异无统计学意义(P>0.05);42 d实验组和28 d实验组相比肺泡上皮细胞Occludin表达明显降低,P<0.05,ZO-1表达差异无统计学意义(P>0.05).结论 COPD时,紧密连接蛋白表达减少,并随时间的延长,Occludin表达进一步降低,由此证明,Occludin和Zo-1表达与肺泡上皮屏障的破坏密切相关.
Objective To explore the potential mechanism of alveolar epithelial structure alternation in COPD. Method We established a COPD rat model, and dissected the rats in batches after 28 days and 42 days from the day establishing the model, then observed the distribution and expression of Occludin and ZO-1 in the rat alveolar epithelium tight junction regions by immunohistochemistry. Results The expression of Occludin and Zo-1 in alveolar epithelia cells of COPD rats declined dramatically (P 〈 0.05) on day 42, the expression of Occludin and Zo-1 in alveolar epithelia cells had a little decrease, but there was no significant difference (P 〉 0.05). Compared to the experimental group on 28 day, the expression of Occludin and Zo-1 in alveolar epithelial cells on day 42 had a dramatic decrease (P 〈0.05), but the expression of ZO-1 had no significant difference (P 〉 0.05). Conclusion In COPD rat model, the expression of tight junction protein decreases, and with the extension of time, the expression of Occludin will decline further more, proving that the expression of Occludin and ZO-1 is closely related to the destruction of alveolar epithelial barrier.
