摘要
在大鼠小肠缺血再灌注 (IIR)模型上 ,研究磷脂酶A2 (PLA2 )在IIR所致肺损伤发病过程中的作用 ,观察大黄对PLA2 影响 ,探讨大黄防治肠源性肺损伤的机理。方法 SD大鼠随机分为肠缺血再灌注组、假手术组、大黄治疗组和安慰剂组。以12 5I标记小牛血清白蛋白 (BSA)肺摄取指数作为评价肺损伤的指标 ,以髓过氧化物酶 (MPO)作为评价多聚核白细胞(PMN)在组织中聚集的指标 ,分别测定各组动物不同时间血清、肺及小肠组织PLA2 活性。结果 大黄可明显抑制再灌注导致的肺MPO活性升高 (P <0 0 1)及肠缺血期和再灌注期血清、肺及小肠组织PLA2 活性升高 (P <0 0 5或P <0 0 1) ,降低肺毛细血管通透性 (P <0 0 1)。结论 早期应用大黄能明显防治大鼠IIR所致的肺损伤 ,这种作用至少部分是通过抑制PLA2
Objectives To evaluate the possible role of phospholipase A 2(PLA 2) in the development of lung injury after intestinal ischemia-reperfusion (IIR),and the effect of rhubarb on PLA 2 on the model of rat intestinal ischemia (45 minutes of superior mesenteric artery occlusion alone) followed by 6 hours of reperfusion.Methods Adult Sprague-Dawley rats were divided into four groups:(1) ischemia and reperfusion alone (n=24),(2) those animals receiving rhubarb 600 mg/kg 30 minutes before reperfusion (n=24);(3) those animals receiving NS instead of rhubarb,serving as placebo (n=16);(4) the remaining 16 animals were sham operated (laparotomy),serving as controls.Lung permeability was measured using bovine serum albumin (BSA) labeled with 125 I,the activity of lung myeloperoxidase (MPO) was measured as an index of polymorphonuclear (PMN) presence.The activity of PLA 2 of serum,small intestine and lung in different groups and in different time was measured.Results Rhubarb down regulated the activity of MPO in the developmennt of lung injury after IIR ( P <0 01),inhibited the activity of PLA 2 in the stage of ischemia and reperfusion,meanwhile the pulmonary microvascular permeability was decreased significantly ( P <0 01) Conclusion The result of this study show that using rhubarb in early stage may prevent lung injury of rats after IIR,this protective effect may at least in part due to the inhibition of PLA 2 [
