摘要
①目的探讨普罗布考在动脉粥样硬化病变中的抗氧化作用及其对增殖细胞核抗原(PCNA)及核因子kappaBp65(NFkB p65)活化的影响。②方法雄性新西兰大耳白兔24只,随机分为正常组、高脂组、普罗布考组,每组8只,共计喂养10周。实验结束后,检测各组家兔的血清甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、超氧化物歧化酶(SOD)以及丙二醛(MDA)水平,并采用免疫组化法测定血管壁PCNA及NFkBp65的表达。③结果与正常组比较,高脂组血清TG、TC、LDL-C明显升高,HDL-C明显降低(P<0.01),与高脂组比较,普罗布考组血清TG降低,但未达到统计学意义(P>0.05),TC、LDL-C明显降低(P<0.01),HDL-C亦有所降低(P<0.05);高脂组较正常组血清SOD明显减少,MDA明显升高(P<0.01),而普罗布考组较高脂组SOD明显升高,MDA明显降低(P<0.01);高脂组较正常组PCNA和NFkBp65表达明显增加(P<0.01),普罗布考组较高脂组PCNA和NFk-Bp65的表达明显减少(P<0.01),相关分析表明PCNA和NFkBp65呈正相关(r=0.916,P<0.01)。④结论普罗布考可减轻脂质过氧化损伤的作用,阻断NFkBp65的活化,进而抑制平滑肌细胞的增殖。
Objective To study the effect of probucol on oxidation and the activity of proliferating cell nuclear antigen (PCAN) and NFkBp65 in atherosclerosis. Methods 24 male New Zealand white rabbits were randomly devided into 3 groups : norreal group, high fat diet group and probucol group. After 10 weeks, the level of serum triglyceride(TG) , total cholesterol (TC) , low density lipoprotein cholesterol ( LDL - C), high density lipoprotein cholesterol (HDL) , superoxide dismutase (SOD) and malondialdehyde (MDA) were measuered. The expression of the PCAN and NFkBp65 in aorta were measured by the method of immu- nohistochemistry. Results Compared with those of the normal group the level of serum TG , TC and LDL - C dramatically increased ( P 〈 0.01 ), but the level of serum HDL - C decreased( P 〈 0.01 ) in the high fat diet group. Compared with that of the high fat diet group the level of serum TG in probucol group decreased ,but had not reached statistical diference( P 〉0.05). In the probucol group, the level of serum TC and LDL - C decreased ( P 〈 0.01 ) and the level of serum HDL - C also decreased ( P 〈 0.05 ) com- pared with those of the high fat diet group. The level of serum SOD was lower and the level of serum MDA was higher in high fat diet group than those of the normal group( P 〈 0.01 ), while the level of serum SOD was higher and the level of serum MDA was lower in the probucol group than those of the high fat diet group( P 〈0.01 ). In the high fat diet group the exepression of PCNA and NFk- Bp65 were advanced compared with those ~of the normal group( P 〈 0.01 ), but they were reduced in the probucol group compared with those of the high fat diet( P 〈0.01 ). The expression of .PCNA waspositively correlated with the expression of the NFkBp65 in the plaque of the aorta( r = 0.916, P 〈 0.01 ).
