摘要
以CEA单克隆抗体C50 包被微孔板 ,异硫氰酸苄基二乙烯三胺四乙酸络合Eu3+ 标记C17单抗。采用平衡法建立CEA时间分辨荧光免疫分析 ,数据采用Log -Logit函数和四参数logit函数数据处理程序处理。方法的批内和批间CV分别为 2 .97%和 1.6 0 % ,平均回收率为10 1.98% ,灵敏度为 0 .2 0 μg/L ,可测范围为 2 .39~ 50 8.9μg/L ,ED50 为 6 0 .91± 0 .56 μg/L。本方法与AFP和CA12 5和CA153无交叉 ,与CA199有 3.2 2 %的交叉反应。Eu3+ 标记抗体于 - 30℃保存一年多 ,免疫反应性基本无损失 ,同批试剂连续 5个月分析结果稳定。与Wallac同类产品比较 ,结果吻合。
A two-site time-resolved fluoroimmunoassay (TRFIA) of carcinoembryonic antigen (CEA) based on the direct sandwich technique has been developed, on the equilibrium method. The monoclonal antibodies (McAb) directed against a specific antigenic site on the CEA was C 50 , the europium-labelled McAb which was prepared by E 3+ u-DTTA directed against a different antigenic site on the CEA molecule was C 17 . The lumineces enhancement system was enhancement solution which contained mainly 2-naphthoyltrifluoroacetone. The intra-and inter-assay CV of the TRFIA were 2.97% and 1.60%, respectively, and the recovery rate was 101.98%, the sensitivity was 0.2μg/L.The McAb provided a linear response from 12.39 to 508.9 μg/L with ED 50 of 60.91±0.56μg/L. The cross reaction with AFP, CA 125 and CA 153 was negligible, and that with CA 199 was 3.22%. The europium-labelled C 17 was stable for a year at -30℃ and the results of the TRFIA with same reagents was stable for five months. Compared with the product of Wallac Cop., their results were same.
