摘要
目的 :为获得编码高亲和力IgE受体(FcεR1)α链的cDNA序列。方法 :从正常人外周血中富集嗜碱性粒细胞 ,提总RNA进行RT PCR ,分子克隆 ,用自动测序及放射自显影测序。结果 :分离并鉴定了该cDNA重组子 ,在159密码子及185密码子分别发现了CAC→CGC与TAT→CAT置换。结论 :建立了一个编码FcεR1膜外区α链的cDNA重组子克隆 ,它不含引导肽序列 ,可能是一个来自中国人的该基因的变异体。
Objective: To obtain the cDNA sequence encoding the alpha subunit of high affinity IgE receptors. Methods: Basophils were enriched in Chinese person. After the total RNA was extracted, RT PCR and cloning were carried out. The sequence was determined with automated DNA sequencing and autoradiography. Results: The cDNA recombinant was sequenced and characterized. It had two transitions at codon 159 CAC→CGC and 185 TAT→CAT. Conclusion: One recombinant was constructed. It contains the cDNA sequence encoding for the extracellular domain of the alpha subunit without the leading peptide sequence. Therefore, it may be a variant from Chinese people.
