陈皮、姜提取物对血瘀证大鼠血液流变学和血小板聚集率的影响 被引量:34

Effects of Pericarpium Citri Reticulatae and Rhizoma Zingiberis extracts on hemarheology and platelet aggregation of blood stasis model rats

摘要 目的:观察陈皮、姜提取物对寒凝血瘀证大鼠血液流变学和血小板聚集率的作用,评价其对血栓形成的预防作用。方法:将60只大鼠随机均匀分为正常对照组、模型对照组、复方丹参滴丸对照药(81mg/kg)、陈皮、姜提取物高(0.36ml/kg)、中(0.18 ml/kg)、低(0.09 ml/kg)剂量组。各组灌胃给药7天,在第6、7两天给药后1h,除空白对照组外,其余各组采用皮下注射肾上腺素(0.8mg/kg)加冰水刺激法复制大鼠血瘀证模型。在第8天末次药后1h采血,测定血液流变学指标和血小板聚集率。结果:与空白组比较,模型组大鼠表现出全血粘度、血浆粘度、红细胞压积、电泳时间、血浆纤维蛋白原明显增加(P<0.01)、红细胞变形性下降(P<0.01)的血液流变学异常现象;血小板聚集率也明显上升(P<0.01);陈皮、姜提取物低剂量对全血高切粘度、红细胞变形性和血浆纤维蛋白原异常的改善作用不明显(P>0.05),其他给药组(0.36、0.18ml/kg)均具有不同程度的改善以上指标的作用,且与复方丹参滴丸没有明显差异(P>0.05)。结论:陈皮、姜提取物具有改善血瘀证大鼠血液流变学异常和抑制血小板聚集,具预防血栓形成的作用。 Objective: To observe the effect of Pericarpium Citri Reticulatae(陈皮) and Rhizoma Zingiberis(姜) extracts(PRE) on hemarheology and platelet aggregation of the blood stasis rats,so as to evaluate the role of it on thrombosis.Methods : 60 rats were randomly divided into normal control group,model control group,positive control group,PRE group(0.36 ml/kg,0.18 ml/kg,0.09 ml/kg),then treated by gavage in consecutive 8 days.At the 6th and the 7th day,All animals except the normal control group were injected subcutaneously adrenaline(0.8mg/kg) and treated with iced water to induce blood stasis.In the 8th day,collected the blood sample from abdominal aorta for the determination of the hemorheology indices and the platelet aggregateon.Results : Compared with the normal control group,the whole blood viscosity,plasma viscosity,hematoctit value,electrophoresis time,plasma fibrinogen of the model control group increased obviously(P 0.01),while the red blood cells deformation de-creased(P 0.01),Platelet aggregation significantly increased(P 0.01) in the blood stasis model rats;compared with the model group,0.09ml/kg PRE group have no obvious difference in the whole blood viscosity,the red blood cells deformation and plasma fibrinogen(P 0.05),other treatment groups all can improve the above indices,and have no significant difference with the positive control group(P 0.05).Conclusion: PRE could improve abnormal Hemarheology and inhibit platelet aggregation of the blood stasis rats,thus,it can prevent thrombosis.
