摘要
目的:建立实验小型猪的冠状动脉内皮细胞体外培养方法。方法:冠状动脉血管块短暂酶消化后采用植块法进行培养。结果:植块培养2日细胞从植块中移出;培养6日植块周围细胞呈铺路石样特征,传代/纯化一次后5日细胞单层贴壁生长,融合成片。细胞因子Ⅷ免疫荧光检测内皮细胞表达阳性,纯度100%。结论:成功建立猪冠状动脉内皮细胞体外培养方法,此方法操作简单、快速,得到细胞纯度高。
To establish the experiment of miniature pigs coronary artery endothelial ceils in vitro culture method. Methods： Coronary arteries after a short piece of enzyme digestion using plant piece of law culture. Results： 2 day, cells removin from plant cell block ing; 6 days, cells are stepping stones to sample around characteristics, ChuanDai/purification 5 days after a single ceils to stick wall growth, conflu- ent. Immune cell factors Ⅷ fluorescence detection endothelial ceils express positive, 100% purity. Conclusion： Building a successful pig coronary artery endothelial ceils in vitro culture method, this method is rapid, simple operation, high purity get cells.
