摘要
目的比较生理盐水稀释法与ARCHITECT HBsAg试剂配套原液稀释法检测HBsAg定量的结果。方法留取HBsAg定量值超过250IU/ml的血清标本共96份,分别用ARCHITECT HBsAg试剂配套原液稀释(A法)、生理盐水稀释(B法)后进行HBsAg定量的检测,将91例均能得到具体测定值的样本纳入统计学分析,比较B法与A法检测结果的一致性。结果 A法、B法测定值的对数值范围分别为(2.72~4.92)lg IU/ml、(2.52~4.97)lg IU/ml,对数值的中位数分别是3.83lg IU/ml、3.81lg IU/ml。B法检测结果与A法检测结果的差异无统计学意义(P=0.712)。B法与A法检测结果的相关系数为0.969(P=0.000)。对检测结果采用Bland-Altman方法分析,结果显示,B法的检测结果与A法的检测结果具有很好的一致性,B法与A法检测结果的差值均数的绝对值接近0,一致性界限范围窄。在不同的HBsAg水平分层中,B法的检测结果与A法亦有很好的相关性。结论 B法的检测结果与A法的检测结果相关性高、差异程度低,具有很好的一致性。
Objective To compare two diluents in the HBsAg quantitative detection.Methods A total of 96 specimens with HBsAg over 250 IU/ml were included in the study.ARCHITECT HBsAg detection kit was used for further dilution analysis to compare two diluents:solution dilution in the kit(A method) and saline dilution(B method).Results Data from 91 cases were included in the statistical analysis.The detection range of methods A and B were(2.72-4.92)and(2.52-4.97)lg IU/ml,respectively;the medians were 3.83 and 3.81 lg IU/ml,respectively.There was no significant difference between diluents A and B(P=0.712).The correlation coefficient of the two methods was 0.969 and with high consistency by Bland-Altman analysis;the mean of difference between method A and B was close to 0.Methods A and B also showed very high correlation at different HBsAg levels.Conclusions Normal saline can be used as the diluents in the detection of HBsAg when using the ARCHITECT kit.
