头痛芷痉提取物镇痛作用研究

摘要 目的观察头痛芷痉提取物的镇痛作用,并初步探讨其作用机制。方法通过小鼠醋酸扭体试验、大鼠硝酸甘油致偏头痛试验及右旋糖苷致家兔血淤试验进行评价。结果头痛芷痉提取物(0.36~1.44 g/kg)连续给药7 d后,可显著延长动物的痛阈值,药后15 min内扭体次数减少,镇痛率升高。头痛芷痉组(0.5~1.0 g/kg)挠头、挠腮、爬笼次数比模型对照组减少,挠头、挠腮出现时间晚于模型组,消失时间则早于模型组,且呈剂量依赖性。头痛芷痉0.448 g/kg可明显降低家兔全血黏度低切、高切和全血还原黏度低切、高切以及血浆黏度。结论头痛芷痉提取物对醋酸溶液引起的小鼠疼痛模型和硝酸甘油引起的大鼠偏头痛模型有较强的镇痛作用;可显著改善血淤模型家兔的血液流变性,具有一定的活血化淤作用。 Objective To observe the analgesic effect and mechanism of headache zhijing extract. Methods The evaluation was performed by acetic acid writhing test in mice, nitroglycerin-induced migraine test in rats and dextraninduced blood stasis test in rabbits. Results After 7 days with the administration of headache zhijing extract (0.36- 1.44g/kg), the animals could significantly prolong the pain threshold, reduce the number of writhing within 15min after administration, and increase analgesic efficacy. Compared with the control group, the number of scratching heads, scratching cheek, climbing the cage were significantly reduced in headache zhijing extract (0.5-1.0g/kg) group, and the appearance of scratching heads and scratching cheek was significantly later and the disappearance was significantly earlier in a dose-dependent manner. Headache zhijing extract (0.448g/kg) could significantly reduce the low and high shearing stress of blood viscosity, and as well as the low and high shearing stress of blood reduced viscosity and plasma viscosity in rabbits. Conclusion Headache zhijing extract has strong analgesic effect on writhing mice induced by acetic acid and migraine rats induced by nitroglycerin, and it can significantly improve the hemorheology in blood stasis model of rabbit.
