摘要 目的:探讨克感复方在MDCK细胞上抗流感病毒的作用并对其机制进行初步研究。方法:以利巴韦林为阳性对照药物,运用抑制试验、预防试验、中和试验3种方法,以MTT法检测细胞存活率,研究克感复方抗流感病毒的作用并判断其作用原理。同时,通过药物血凝抑制实验分析其抗流感病毒作用是否与病毒血凝素相关。结果:克感复方对MDCK细胞的半数毒性浓度(CC50)为(2.84±0.65)mg/mL,抑制流感病毒增殖的半数有效浓度(EC50)为(0.18±0.12)mg/mL,选择性指数(SI)为15.78。有细胞外杀流感病毒作用,但作用较弱,最高抑制率未超过50%。无阻断流感病毒吸附的作用。无药物血凝抑制作用。结论:克感复方在MDCK细胞上具有抗流感病毒的作用,其中抑制流感病毒增殖的能力最强。 Objective:To study the effects of Kegan compound against influenza virus in MDCK cell and preliminary study its mechanism.Methods:Ribavirin as a positive control,use of Post-infection treatment assay,Pre-infection treatment assay,Virucidal assay three methods.By MTT method to detect cell viability,research the activities of kegan compound anti-influenza virus.In addition,hemagglutination inhibition test analysis that drug anti-influenza virus effect whether interaction with influenza virus hemagglutinin.Results:The 50% cytotoxic concentration(CC50) of Kegan compound was 2.84±0.65mg/mL,Post-infection treatment assay measured the 50% effective concentration(EC50) was 0.18±0.12 mg/mL,and the SI was 15.78.Virucidal assay is weak relatively,the maximum inhibition did not exceed 50%.No Pre-infection treatment effect.No hemagglutination inhibition effect.Conclusions:Kegan compound have obviously inhibited effects on influenza virus in MDCK cell,and the effects are strongest on inhibit the virus multiplication.
