摘要
目的:通过胚胎期给药,探讨补肾方对胎儿宫内发育迟缓(IUGR)模型胎鼠己糖激酶(HK)、谷氨酸脱氢酶(GDH)活性的影响。方法:模型组用酒精给孕鼠灌胃导致IUGR模型,各补肾组分别在造模同时予补肾方灌胃,其他组予补气方、补血方、气血双补方灌胃,检测各组胎鼠肝组织HK、GDH活性。结果:IUGR胎鼠HK、GDH活性明显降低(P<0.05);补肾阴方、补血方对IUGR胎鼠HK活性降低均有明显恢复作用(P<0.05);补肾方、补血方、气血双补方均能不同程度恢复IUGR胎鼠GDH活性(P<0.01,P<0.01,P<0.05,P<0.05)。结论:改善IUGR胎鼠肝组织HK、GDH活性可能是补肾方治疗IUGR机理之一,可作为中医"肾主生殖发育、肾为先天之本"和"补母益子"论的依据。
Objective:This research is to explore the effect of prescription tonifying kidney on hexoxinase(HK)and glutamate dehydrogenase(GDH)in liver tissue of fetuses with intrauterine growth retardation(IUGR).Methods:The pregnant mice were randomly divided into 9 groups.The control group were fed normally.And other control group were fed by Zuogui Pill(ZGP,a prescription for reinforcing kidney Yin).The model mice of IUGR was induced by feeding alcohol.In other 6 experiment groups,the pregnant mice were induced by feeding alcohol and respectively by Zuogui Pill or Yougui Pill(YGP,a prescription for reinforcing kidney Yang)or Wuzi Yanzong Pill(WZYZP,a prescription for reinforcing both of kidney yang and kidney yin)or Sijunzi Decoction(SJZD,a prescription for supplementing Qi)or Siwu Decoction(SWD,a prescription for supplementing blood)or Bazhen Decoction(BZD,a prescription for supplementing Qi and blood)at the same time.19 days later,the activities of hexoxinase and glutamate dehydrogenase in liver issue of fetuses were detected.Results:The activity of HK and activity of GDH in liver tissue of mice with IUGR were both lower(P〈0.05).ZGW and SWD can both enhance the activity of HK in liver tissue of mice with IUGR(P〈0.05).ZGP,YGP,SWD and BZD all can enhance the activity of GDH in liver tissue of mice with IUGR(P〈0.01,P〈0.01,P〈0.05,P〈0.05).Conclusion:One mechanism of prescription for tonifying kidney is to enhance the activity of HK and activity of GDH in liver issue of mice with IUGR.And this may be an evidence of the theories of kidney controlling reproduction and upgrowth and invigorating the mother to benefit the children in TCM.
