摘要
建立了荷移荧光光谱法测定杨树桑黄提取物中总黄酮含量的方法。以芦丁为对照品,1%三氯化铝为受电子试剂,测定荷移复合物荧光强度,并系统考察了荷移反应条件对荧光强度的影响。芦丁在4—20μg·mL^(-1)浓度范围内线性关系良好,RSD为2.21%,平均回收率为100.3%。荷移反应后黄酮类物质产生荧光,可用于定量测定,方法简便、快速,适用于杨树桑黄总黄酮的含量测定。
The method for determination of total flavonoids in Phellinus vaninii extract by spectrofluorimetry after charge transfer reaction was established.Rutin was chosen as the reference substance and 1%aluminum chloride was chosen as the electron acceptor reagent in charge transfer reaction.Fluorescence intensity of charge transfer complex was detected,on which the effects of charge transfer reaction conditions were investigated.The content of total flavonoids in Phellinus vaninii extract was determined by spectrofluorimetry.The calibration curve was linear in the range of 4-20μg·mL-1 for rutin with RSD of 2.21%and the average recovery was 100.3%.After charge transfer reaction flavonoids can generate fluorescence,which can be used in quantitative determination.The method is simple,rapid and can be used for detecting the total flavonoids in Phellinus vanini.
