凋膜止崩方对无排卵功血大鼠增生子宫内膜中Bcl-2和Bax蛋白表达的影响 被引量:2

The Influence of Diao-mo-zhi-beng Prescription on Expression of Bcl-2 and Bax in Hyperplastic Endometrium of Rats

摘要 目的:探讨凋膜止崩方宫腔靶向干预前后Bcl-2和Bax在无排卵功血(ADUB)大鼠增生子宫内膜组织中的蛋白表达情况。方法:将健康成年雌性SD大鼠40只,随机分成空白组(A)、模型组(B)、实验大剂量组(C)、实验小剂量组(D),去势大鼠雌激素负荷造模成功后宫腔注药,采用West-ern-blot方法检测4组大鼠子宫内膜组织中Bcl-2和Bax蛋白的表达。结果:动情前期A组Bcl-2表达较低,B组表达较高;且A组与B组、B组与C、D组之间差异有统计学意义(P<0.05)。而动情前期A组Bax表达较高,B组表达则较低;且A组与B组、B组与D组之间差异有统计学意义(P<0.05)。结论:凋膜止崩方可能通过下调ADUB大鼠增生子宫内膜中Bcl-2蛋白表达,上调Bax蛋白表达,加速子宫内膜细胞凋亡,达到祛除子宫内膜的目的。 Objective: To explore the modification of Bcl-2 and Bax before and after targeting interventions of Diao-mo-zhi-beng Prescription and the expression of protein in the endometrium hyperplasia of rat uterine tissue. Methods: Healthy adult female SD rats were randomly divided into 40, the control group ( A ), model group ( B ), large dose of Diao-mo-zhi-beng prescription ( C ), small dose of Diao-mo-zhi-beng prescription ( D ), castrated rat combine estrogen load modeling success of intrauterine injection of medicine, The protein expression levels of Bcl-2 and bax were checked by Western-blot. Results: In preoestrus, the expression of Bcl-2 in group A is lower and the expression of Bcl-2 in group B is higher. For expression of Bcl-2, group A has significant difference with group B ( P 〈 0. 05), and group B has significant difference with group C and group D, respectively ( P 〈 0. 05 ) . And In preo- estrus, the expression of Bax in group A is the higher and the expression of Bax in group B is the lower. For expression of Bax, group A has significant difference with group B ( P 〈 0. 05 ), and group B has significant difference with group D ( P 〈 0. 05) . The expres- sion of Bcl-2 has negative correlation with the expression of Bax in both group A and group B. Conclusion: The diaomozhipen prescrip- tion can dispel the extreme hyperplastic endometrium of rats, and weaken the expression of Bcl-2 while enhance the expression of Bax. Moreover, the apoptosis of endometrium cell is accelerated.
