摘要
目的:研究通天草提取物对SD大鼠局灶性脑缺血(MCAO)模型的影响,探讨其在治疗脑缺血疾病方面的应用价值。方法:将75只健康雄性SD大鼠随机分为空白对照组、模型组、水提组、醇提组和中药对照组。动物于造模后第4天开始给药,连续给药21天后,取血、灌注取脑。检测凝血酶原时间(PT)及脑组织结构。结果:醇提组与模型组及空白组比较,PT值显著增大;光镜下,模型组大面积脑组织坏死及炎性细胞浸润,胶质细胞及血管水肿,多数神经元固缩、坏死;醇提组可见小面积的组织坏死及炎性细胞浸润,程度较模型组轻,偶见血管及胶质细胞水肿。中药组及水提组与模型组及空白组比较,未见显著差异。结论:通天草醇提物可显著抑制MCAO模型大鼠的血栓形成和改善脑组织损伤。
Objective :To study the effects of extracts from watemut herb on MCAO rats and explore its application value in the treatment of cerebral ischemia disease. Methods : 75 healthy male SD rats were randomly divided into five groups : con- trol group, models group, water extraction group, alcohol group and control group. Rats were administrated from the 5th after modeling, collecting blood and brain after continuous administration for 21 days. Detecting PT and brain structure. Results : PT was significantly increased when alcohol group compared with control group and model group ; Under light mi- croscope, large area of brain tissue necrosis, inflammatory cell infiltration, glial cells and vascular edema, and most neu- ronal pyknosis, necrosis were seen in model group; Small area of tissue necrosis, inflammatory cell infiltration, occa- sional blood vessels and glial cell edema could be seen in alcohol group;Water extraction group and control group com- pared with control group and model group were with no significant difference. Conclusion:Waternut Herb ethanol extracts significantly inhibit the MCAO rat models of thrombosis and improve the condition of brain damage.
