摘要 以鼠李糖乳杆菌(ATTC 53103)为出发菌株,通过单因素试验和正交实验筛选优化基础培养基:12%脱脂乳粉、10%的葡萄糖、1%的酵母粉、2%的酪蛋白水解物、0.15%的吐温80。10 L自动发酵罐补料高密度培养条件为:葡萄糖浓度低于4 g.L-1时补加葡萄糖,使罐内葡萄糖浓度稳定在4~6 g.L-1之间,在此条件下发酵20 h,最大活菌数可达到3.75×1010cfu.mL-1,而发酵罐分批培养最大活菌浓度仅为3.8×109cfu.mL-1,前者比后者提高近10倍。 In this study, Lactobacillus rhamnosus (ATTC 53103) was used as the starting strain. Through single factor experiments and orthogonal design, the optimal basic culture medium was selected as the followings 12% skim milk powder, 10% glucose, 1% yeast extract, 2% casein hydrolyzate, 0.15% Tween 80. Cultivating condition in 10 L automatic fermentor, adding glucose from the concentration of 4-6 g. L1 were best for Lactobacillus rhamnosus (ATTC 53103) fed-batch high-densityculture. As mentioned above, the strains were in the logarithmic phase during 20 h, the max viable count could reach 3.75×10^10 cfu·mL-1 at the end of logarithmic phase. However, the maxviable count was only 3.8×10^9 cfu·mL-1 via automatic fermentor batch culture, the former increased by 10 times.
