大鼠脑损伤时神经细胞c-fos、c-jun和c-myc基因表达变化 被引量:6

摘要 目的观察大鼠脑损伤后不同时间皮质神经细胞早期快反应基因c-fos、c-jun和c-myc表达变化。方法Northern杂交、原位杂交和免疫组织化学方法检测皮质神经细胞中c-fos、c-jun和c-myc的表达。结果脑损伤后15min,伤侧皮质神经细胞出现上述3种基因mRNA的表达,随时间的处长,表达逐渐增加,于伤后3h达高峰并持续至伤后24h,伤后72h恢复至对照水平。c-fos和c-jun蛋白表达亦于伤后15min出现,c-myc蛋白表达于伤后30min出现,并均于伤后3h达高峰,持续至伤后6h,伤后24h表达消失。结论脑损伤后上述3种基因表达特点与其它脑损害不同,可能与损伤机制和继发性神经细胞损害有关。 Objective To observe the expression of c-fos, c-jun and c-myc genes in cerebral cortical neurons following brain trauma in rats, Methods The expression of c-fos. c-jun and c-myc was detected by using Northern and in situ hybridization and immunocytochemistry Results The expression of the mRNA of c-fos, c-jun and c-myc genes was observed in neurons 15 min following the injury in the ipsilateral cerebral cortex, which increased gradually and reached their peak at h 6. lasting till h 24 and failed to be detected at h 72, c-fos and c-jun proteins expression were present 15 min after injury and c-myc protein after 30 min. peaking at h 3 and lasting till h6. and resumed normal level at h 24. Conclusions The expression characteristic of c-fos. c-jun and c-myc gene differs from that in other brain injures. which may be related to different injury mechanism and secondary neuronal damage,
