摘要
以呋喃唑酮代谢物(AOZ)和卵清蛋白(OVA)偶联物为包被抗原,与AOZ样品或标准品竞争呋喃唑酮多克隆抗体,加入辣根过氧化物酶(HRP)标记羊抗兔抗体(IgG),研制AOZ的化学发光酶联免疫检测试剂盒。以鲁米诺-过氧化氢为发光底物进行化学发光酶联免疫检测。结果表明:试剂盒分析灵敏度为0.005μg/L,比传统的ELISA试剂盒高出一个数量级,拥有更低的检测限,批内变异系数为2.1%~7.9%,批间变异系数为5.2%~9.5%,批内回收率为93%~113%,批间回收率为94%~110%。与呋喃西林代谢物(SEM)、呋喃它酮代谢物(AMOZ)及呋喃妥因代谢物(AHD)之间无明显交叉反应。此试剂盒可用于水产品中AOZ的检测。
Taking AOZ-VOA as enveloped antigen, which competes with AOZ samples or standard AOZ to get furazolidone polyclonal antibody, and joining HRP to label IgG. a chemiluminescsences-ELISA kit for the monitoring of AOZ residue is developed. Tests ELISA taking luminol-H2O2 as substrate show that the sensitivity of the AOZ CL-ELISA kit is 0. 005 μg/L,one order higher than that of conventional ELISA kits, and has a lower detection limit, and that the inter CV is 2.1% - 7.9%, the intra CV 5.2% - 9.5% ,the inter analytical recovery 93% - 113%, and the intra analytical recovery 94% - 110%. This kit has no visible cross reaction with SEM, AMOZ and AHD, so it can be used to detect AOZ in aquatic products.
