摘要
目的应用薄层色谱、高效液相色谱-二极管阵列检测和气质联用技术定性定量分析胆安合剂(大黄、元胡、黄芩、厚朴、木香、金钱草、甘草等)中的药效物质。方法应用反相C18色谱柱,甲醇-0.1%磷酸(88∶12)为流动相,体积流量0.8 mL/min,紫外检测波长209 nm,220 nm,柱温40℃进行高效液相色谱分析;高纯氦气为载气,体积流量1 mL/min,EI离子源进行气质联用分析。结果胆安合剂提取物薄层层析有明显的特征斑点;延胡索乙素、厚朴酚、大黄素、大黄酚的质量浓度分别为10.68～10.88μg/mL,12.25～12.98μg/mL,2.56～2.71μg/mL,3.54～3.67μg/mL;气质联用分析鉴定相似比≥800的挥发性化合物54个。结论己醛、3,7-二甲基-1,6-辛二烯-3-醇、4-甲基-1-(1-异丙基)-3-环己烯-1-醇(R)、薄荷基-1-烯-8-醇、桉醇基-4(14)-烯-11-醇和巴西菊内酯6种挥发性物质可作为总挥发油的特征成分加以控制。
AIM To analyze effective constituents in Dan＇an Mixture（Rhei Radix et Rhizoma,Corydalis Rhizoma,Scutellariae Radix,Magnoliae officinalis Cortex,Aucklandiae Radix,Lysimachiae Herba,Glycyrrhizae Radix et Rhizoma,etc.） by TLC,HPLC-PDA and GC-MS methods.METHODS The HPLC analysis was performed on a C18 silica column by using methanol-0.1% phosphoric acid（88∶12） as mobile phase at a flow rate of 0.8 mL/min.UV detection wavelengthes were set at 209 nm and 220 nm,column temperature was maintained at 40 ℃.High purity helium was used as the carrier gas at a flow rate of 1 mL/min for GC-MS analysis with electron impact source.RESULTS The extracts of Dan＇an Mixture had characteristic spots in TLC analysis.The contents of tetrahydropalmatine,magnolol,emodin and chrysophanol were in the ranges of 10.68-10.88 μg/mL,12.25-12.98 μg/mL,2.56-2.71 μg/mL and 3.54-3.67 μg/mL,respectively.Fifty-four volatile compounds were obtained with similitude ratio greater than 800.CONCLUSION Hexanal,3,7-dimethyl-1,6-octadien-3-ol,4-methy-1-（1-methylethyl）-3-cyclohexen-l-ol（R）,ρ-menth-l-en-8-ol,encalyptoly-4（14）-ene-11-ol and eremanthin can be considered as the characteristic indexes of volatile oil for quality control.
