产葡萄糖氧化酶工程菌培养基筛选及发酵条件优化 被引量:2

摘要 利用单因素试验和4因素3水平的正交试验L9(34)对1株产葡萄糖氧化酶毕赤酵母工程菌HL-69进行了研究,确定了该菌株的适宜廉价生长培养基和诱导培养基。生长培养基:葡萄糖1.5%、豆粕粉3.0%、磷酸二氢钾0.2%、硫酸镁0.03%;诱导培养基:甲醇1.5%、氨水3.5%、磷酸二氢钾0.5%、硫酸镁0.03%。培养条件为:接种龄18h,接种量2%,培养基装液量为50mL/300mL,生长初始pH为5.0,诱导初始pH为6.0;在此条件下发酵,葡萄糖氧化酶酶活力达6.516U/mL,是发酵条件优化前酶活力的7.50倍。 The medium and fermentation conditions were studied via Single factor experiment and orthogonal test(four factors and three levels) to a stain producing glucose oxidase. The optimized growth medium and fermentation medium were respectively as follows:growth medium: glucose 1. 5%,soypeptone 3.0%,KHzPO40.2 %,MgSO4 · 7H2O 0. 03%;fermentation methanol medium: 1.5% ,ammonia 3.5% ,KH2PO40.5% ,MgSO4 · 7H2O 0.03%. The best fermentation conditions were as follows: inoculating time 18 h, inoculating amount 2%,medium volume 50 mL in 300 mL flask,initial growth medium pH 5.0 and initial fermentation medium pH 6.0; Under this condition, the activity of glucose oxidase is 6. 516U/mL, which is 7. 50 times of the initial strain.
