摘要
目的观察脓毒症小鼠脾淋巴细胞凋亡情况。方法采用盲肠结扎穿孔术复制小鼠脓毒症模型,30只小鼠随机分为假手术组(Sham组)和模型组(CLP组),术后24h取脾组织,行HE染色,观察病理变化,TUNEL法检测细胞凋亡。结果 CLP组脾脏白髓淋巴滤泡内则见大量灶性分布的凋亡淋巴细胞,凋亡的严重程度以凋亡指数(Apoptosis Index,AI)计算,CLP组与Sham组比较差异具有统计学意义(P<0.01)。结论脓毒症脾淋巴细胞凋亡明显增加,成为脓毒症免疫抑制的重要机制。
Objective To investigate whether there was extensive apoptosis of lymphocytes in the clini- cally relevant model of sepsis. Methods This study was performed cecal ligation and puncture (CLP) on mouse to induce sepsis and observed consecutively the effect of splenic tissue. 30 mice were assigned ran- domly to two groups: sham operated group (Sham) , cecal ligation and puncture group (CLP) ,each of 15. Twenty-four hours after surgery, mice per experimental group were sacrificed,spleen was removed used for hematoxylin and eosin, apoptosis cell was detected in TUNEL. Results The changes of splenic lym- phocyte apoptosis.. In the CLP group there were a great quantity, and distributed in focus. The serious of apoptosis was remarked with apoptotic index. Comparison between sham and CLP group, the difference had statistical significance(P〈0.01). Conclusion Sepsis induces extensive lymphocyte apoptosis in the spleen. So lymphocyte apoptosis is an important mechanism of immune suppress in sepsis.
