摘要
目的:观察在旋肉碱及缺血预处理(ischemicpreconditiong)对家兔缺血—再灌注所致心肌细胞凋亡的影响。方法:21只成年大白兔被随机分为3组:对照组、肉碱组和缺血预处理组。3组家兔均经历20分钟冠状动脉左前降支结扎,再灌注4小时。在此之前.以灌胃方式给予肉碱组家兔6天左旋肉碱;缺血预处理组则于缺血前20分钟经历3次短期缺血—再灌注。再灌注末取缺血区心肌组织切片缺口末端标记法显示凋亡细胞核,光镜观察。结果:对照组心肌细胞凋亡率与缺血预处理组比较无差异(对照组:15.16%±9.8%;缺血须处理组:14.77%±3.8%,P>0.05);肉碱组心肌细胞凋亡率则明显少于以上2组(肉碱组:3.24%±6.49%,P<0.05。结论:主旋肉碱能明显阻止缺血—再灌注所致心肌细胞凋亡的发生,缺血预处理不能阻止缺血—再灌注所致心肌细胞凋亡的发生。
Objective: To investigate the effect of L-carnitine against myocardial ischemia-reperfusion induced apoptosis of rabbit heart and to compare its effect with that of ischemic preconditioning. Methods: Twenty-one adult white rabbits were equally divided into three groups: control group, carnitine group and preconditioning group. All rabbits underwent 20 minutes of left anterior decending coronary artery ligation followed by 4 hour reperfusion. Before this procedure, the carnitine group were given levocarnitine for 6 days (40 ng· kg· d(-1); the preconditioning group underwent 3 cycles of short period of ischemia and reperfusion just before the procedure. Afer the reperfusion period, myocardial cell apoptosis were detected using TUNEL method. Results: The rate of apoptotic cell of the control group and the preconditioning group was not significantly different (control) group: 15. 16% ± 9.8%, preconditioning group: 14.77%± 3 .8%, P > 0.05), but the rate of apoptotic cell of the carnitine group was significantly lower than that of the other two groups. Conclusions: Levocarnitine had a protective effect against ischemia- reperfusion induced apoptosis of the rabbit heart, ischemic preconditioning did not have this kind of effect.
