摘要
目的探讨结直肠癌及人结肠癌细胞系SW620中CD44+/Ki-67-癌细胞的形态特点、数量及分布特征。方法利用组织芯片技术、免疫组化染色、免疫组化双重染色和HE染色,连续切片,同部位观察。结果正常肠黏膜中CD44蛋白不表达,Ki-67蛋白低表达;腺瘤中CD44蛋白低表达,Ki-67蛋白表达率为5.06%,CD44+/Ki-67-细胞数为0.58%。结直肠癌中CD44+/Ki-67-癌细胞数平均为5.82%,主要分布在腺体基膜缘或腺体共壁侧,分布有统计学意义(P<0.05)。癌细胞主要有两种形态,一种是大核细胞,另一种细胞核较小,形似淋巴细胞;在SW620中CD44+/Ki-67-主要表达在小圆细胞和单极细胞中,其它细胞中表达少;细胞数比率与组织学相比,差异无显著性(P>0.05)且比值接近;Ki-67-的癌细胞中,CD44+癌细胞占67%,CD44-癌细胞占33%,两者差异有显著性(P<0.05),说明大多CD44+癌细胞处于G0期或静息期,只有少数细胞处于细胞G1、S、G2、M期。结论 CD44+/Ki-67-癌细胞特征和从癌干细胞理论方面考虑,更适合作为癌干细胞的标记,为癌干细胞的分离、靶向治疗和个体化治疗提供可靠的分子生物学指标。
Purpose To explore the cell characteristics, quantity and distribution of the CIM4^+/Ki-67^- in human colorectal cancer cell lines SW620 and the colorectal cancer. Methods Using tissue chip technology and immunohistochemical staining, double immu- nohistochemical staining and hematoxylin-eosin staining, the changes in the same part of a continuous paraffin section were observed. Results In normal mucosa, the expression of CD44 protein was negative, and Ki-67 was of low rate. In adenoma, the expression of CD44 protein was low, and Ki-67 protein was 5.06% , and the CIM4^+/Ki-67^ - cells was in an average of 0. 58%. In colorectal cancer tissue, the CD44 ^+/Ki-67^- cancer cells was in an average of 5.82% and mainly distributed along the side basement of the gland or be- tween the glands, with significant difference ( P 〈 0. 05 ) . There were basically two kinds of cells : one with a larger nucleus, and an- other with small nuclei, similar to a lymphocytes. In SW620, the ratio of CD44^+/Ki-67^ - cell was close to the colorectal cancer ( P 〉 0.05 ) , and the positive expression was mainly detected in the small round cells and unipolar ceils, but other ceils less expressed. In Ki-67 -cancer cell group, the CD44^+cancer cells accounted for 67% and the CD44^+ cancer cells accounted for 33% (P 〈 0. 05 ). This proved that most CD44^+ cancer cells were in the Go or static phase, and only a few cells were in the G1 , S, G2 and M phases. Conclusions Based on the cell features and the theory of cancer stem cells, CD44 ^+/Ki-67^ - cancer cells are more suitable as a mark- er of cancer stem cells. It may provide a reliable molecular biology parameter for the isolation of cancer stem cells and targeted therapy.
