摘要
目的建立绿脓杆菌(PA,又名铜绿假单胞菌)肺炎动物模型,并从病理学、微生物学、影像学方面进行评价,探索并优化建模方法,为临床绿脓杆菌肺炎相关研究提供可行的动物模型基础。方法 24只健康清洁级新西兰大白兔随机分成2组,实验组采用喷雾吸入法,隔日接种反复感染家兔,对照组予以等量生理盐水。接种后隔日一次行胸部CT扫描。比较二组家兔的体重、体温、血象及精神状态;观察实验组病理组织学和影像学改变。结果吸菌组家兔体重下降,白细胞总数显著升高,早期出现寒颤发热,躁动不安,晚期反应迟钝。大体标本早期有明显的充血、水肿及颜色变化等改变,进展期出现脓肿、出血、实变、质地变硬等。镜下观察表现为急性、慢性肺炎和急慢性混杂的病理组织学改变。结论使用喷雾吸入法反复感染新西兰兔,可成功建立家兔绿脓杆菌肺炎模型。
Objective To establish an animal model of Pseudomonas aeruginosa (PA) pneumonia while exploring and optimizing modeling approach, and to evaluate the animal model with imaging, pathological, microbiological studies. Methods 24 healthy, clean New Zealand rabbits were randomly divided into 2 groups: i.e. group A were inoculated with PA suspen- sion by inhaling every other day and Group B inhaled saline suspension as the control group. Body weight, temperature, blood and spirit were compared between these two groups; pathological and radiological changes were observed in group A, Results Compared with group B, the white blood cells(WBC) were increased and weight lossed. Obvious congestion, edema and color changes were found on lung specimens in early period, then abscess, hemonhage, consolidation, texture hardens and so on in advanced period. Acutechronic mixed pathological changes could be seen under microscope. Conclusion The model of Pseudomonas aeruginosa pneumonia can be established by inhalation with PA suspension repeatedly in rabbits.
