低分子肝素对大鼠大脑皮层神经细胞缺血再灌注损伤的保护作用 被引量:1

摘要 目的研究低分子肝素(low molecular weight heparin,LMWH)对大鼠大脑皮层神经细胞缺血再灌注损伤的保护作用及其可能机制。方法体外培养新生大鼠大脑皮层神经细胞,建立缺血再灌注模型,MTT法检测细胞活力,Annexin V-FITC、PI双染流式测细胞凋亡率,荧光分光光度计法测定细胞内钙离子浓度。结果低分子肝素可提高缺血再灌注损伤的神经细胞活力,降低细胞凋亡率和细胞内钙离子浓度。结论低分子肝素对缺血再灌注损伤的大鼠大脑皮层神经细胞有保护作用,其机制可能与LMWH降低细胞内钙离子浓度有关。 Objective To study the protective effects and the mechanisms of low molecular weight hepa rin (LMWH) on ischemiareperfusion injury in rat cerebral cortical neurons. Methods Neonatal rat cerebral cortical neurons were cultured in vitro and the ischemiareperfusion models were established. Neurons viabilities were detected by MTT and Annexin VFITC,PI double staining flow cytometry were used to assess cell apoto sis. Fluorescence spectrophotometer was used to calculate the the intracellular calcium ion concentration in neurons. Results LMWH can improve the vitalities of neurons damaged by ischemiareperfusion injury,and re duce the rate of apoptosis and the intracellular calcium concentration in damaged neurons. Conclusions LMWH have protective effects on ischemiareperfusion iniury in rat cerebral cortical neurons. The mechanisms may be associated with the reduction of the intracellular calcium concentration in neurons caused by LMWH.
