摘要
目的:探讨银杏叶提取物(GBE)对高同型半胱氨酸血症(HHcy)血管内皮受损的保护作用。方法:51只雄性Wistar大鼠随机分为4组,空白对照组12只,模型组13只,叶酸组13只及银杏叶组13只。参照王禄增及Wang G P等造成大鼠HHcy模型。空白对照组予1%CMC液2 mL灌胃,模型组予3%蛋氨酸混悬液1.5 mg/g体重灌胃,叶酸组予3%蛋氨酸混悬液,加叶酸混悬液0.06 mg/g体重灌胃,银杏叶组予3%蛋氨酸混悬液加银杏叶混悬液0.02 mg/g体重灌胃。每日1次,然后给予正常饲料,共8 w。8 w后采用液相色谱-串联质谱(LC-MS/MS)法测定血清Hcy水平,同时测定血浆vWF、FIB、Ps浓度。结果:模型组血清Hcy水平,血浆vWF、Ps、FIB水平明显高于空白对照组(P<0.01)。用药后叶酸组、银杏叶组血清Hcy水平,血浆vWF、Ps、FIB水平较模型组明显降低,比较有统计学意义(P<0.01,P<0.05)。结论:GBE可能通过降低HHcy血浆vWF、Ps、FIB水平,减少血栓形成,改善血管内皮功能,延缓HHcy动脉粥样硬化的进程。
Objective:To investigate protective effect of Gingko Biloba extracts( GBE )on endothelial injury in hyperhomoc- ysteinemia rats. Methods:51 male Wistar rats were randomly divided into 4 groups:blank control group, model group, folic group and BGE group. 8 weeks after administration,level of homocysteine (HCY)in serum was detected by LC-MS/MS; concentrations of yon Willebrand Factor(vWF), P-selection(Ps),fibrinogen(FIB)in plasma were detected. Results:Compared with blank control group,levels of HCY in serum, concentrations of vWF, Ps, FIB in plasma were increased significantly in model group (P〈0.01). Compared with model group,levels of HCY in serum,concentrations of vWF,Ps,FIB in plasma were decreased significantly in folic group and GBE group (P〈0.05,P〈0.01), Conclusion :GBE can reduce the levels of HCY in serum,concentrations of vWF,Ps and FIB in plasma,which might be one of mechanisms for delaying atherosclerosis in HHcy.
